Graphs OTRs in 96-squarewell MTPs
The graphs below show the dependence of oxygen-transfer rates (OTRs) in cultures in 96-square deepwell MTPs shaken on a shaker with a 50 mm shaking amplitude, at various shaking frequencies and culture volumes. The kLa values are calculated from the OTRs, assuming a maximal concentration of O2 in the medium of 0.2 mmol per liter.
The data are based on the following publications :
- Duetz, W.A., Rüedi, L., Hermann, R. O'Connor, K, Büchs, J., and Witholt, B. (2000). Methods for intense aeration, growth, storage, and replication of bacterial strains in microtiter plates. Applied and Environmental Microbiology 66:2641-2646
- Duetz, WA, Witholt, B. (2004). Oxygen transfer by orbital shaking of square vessels and deepwell microtiter plates of various dimensions. Biochemical Engineering Journal 17: 181–185
Most data points in the graphs are interpolations and extrapolations of the data in these publications, taking into account the smaller or larger surface-to-volume ratios, as measured from stills from our collection of slowmotion videos.
The graphs also include information of the maximal culture volume where the culture is still fully mixed ("down-to-the-bottom"). Above this critical culture volume, heavy and large cells (such as CHO- or HEK cells, and yeast cells) may precipitate at the bottom (and lead to cell death).
